FOXO4
FOXO4-DRI is a retro-inverso D-amino acid peptide whose sequence recapitulates the FOXO4 domain responsible for anchoring p53 inside senescent cells. When introduced into culture, it outcompetes the endogenous FOXO4-p53 interaction, rerouting p53 toward pro-apoptotic activity and triggering cell death preferentially in p21-high senescent populations. The selectivity for senescent over proliferating cells has made it a widely adopted tool compound for dissecting senolytic mechanisms and evaluating cellular senescence pathway pharmacology in preclinical models.
